Could be a lot of things......
Obviously you can still connect to the internet.......can all (how many are there?) PC's connect to web, or are they just not connecting with each other?

What firewall/AV software?

Do you use a hub/switch/router or direct connection?

Proxy software or not?

Just occasionally, one of the 3 other pC's i've got on this network resets it's IP addy from 192.168.0. blah.....to something completely different, for no reason that I've ever been able to work out. Check all the IP addys are as they should be.

Make sure no one has inadvertently disabled "file sharing" ...had a prob with that too once.

Can't think of anything else at the mo'........i'm assuming you've switched off all pC's then back on again.

If you have widows xp try to disaple the xp´s own firewall if not disapled.
I had a lot of problems after i disapled it in my computer, it allowed me to go internet but sometimes it just stopped downloading and it didnt allow me to contact other computer in lan....and i couln´t sent enything in irc.

All those were fixed when i change it.
Hope it would be your problem too....easy to fix :)

Drop to a command prompt if it happens again, type:

ipconfig /all

and take a look at your IP address if it begins with 169.254 then youve lost your internet connection and have been unable to get an IP address and will have to talk to your isp.

Most reasons for loss of connectivity is this, also spyware is a pain in the a$$ so try and get hold of a spy removal proggy.

Just my 2 euros worth.
